Biological Activity
SARS-CoV-2 nsp3-IN-3 (compound 26a) at concentrations of 200-2000 μM potently inhibits the recombinant SARS-CoV-2 Nsp3 macrodomain Mac1 in an HTRF-based in vitro peptide displacement assay, with an IC50 of 7.24 μM and a ligand efficiency of 0.40[1].
SARS-CoV-2 nsp3-IN-3 (100 mM; 1 h) binds to the active site of the recombinant SARS-CoV-2 Nsp3 macrodomain Mac1, forming specific hydrogen bonds with the Asp22, Phe156 and Asp157 residues, and this finding is confirmed by X-ray crystallography[1].
SARS-CoV-2 nsp3-IN-3 does not inhibit the hydrolytic activity of recombinant SARS-CoV-2 Nsp3 macrodomain Mac1 even at a concentration as high as 1 mM in the AMP-Glo assay[1].
