18:1 DGS-NTA(Ni) is a nickel chelating lipid. 18:1 DGS-NTA(Ni) has high affinity for histidine-tags and can bind histidine tags of recombinant proteins. 18:1 DGS-NTA(Ni) can be used for preparation of liposomes and nanosize multilamellar vesicles (NMVs) which are used in protein and peptide binding studies and antigen delivery.

In Vitro
18:1 DGS-NTA(Ni) (15% of the total mass of DOPC:Cholesterol; 2-18 h) enables surface coupling of histidine-tagged rStx2B to NMVs and supports gradual antigen release while preserving antigenic integrity[1].

Animal Model:BALB/c (female, 6-8 weeks old, challenged with intravenous 1 LD100 of Stx2a)[1]

Dosage:15% of total DOPC:Cholesterol mass (as part of NMVs loaded with 25 μg rStx2B)

Administration:subcutaneous; three times with 2-week intervals

Result:Increased anti-rStx2B serum IgG titers up to tenfold compared to rStx2B alone.
Achieved an Stx2-neutralization IC50 value of 365.7, significantly higher than the IC50 of 62.72 from rStx2B-only immunized mice.
Induced a mixed Th1/Th2 response with detectable IgG1 and IgG2a subclass titers.
Enabled 60% of immunized mice to survive the lethal Stx2a challenge, while 0% of rStx2B-only or non-immunized mice survived past day 3.
