In Vitro
(2S,3aS,7aS)-octahydroindole‑2‑carboxylic acid (Oic), a proline analog, exhibits excellent trans-amide stability and a strong n→π* orientation. It forms a stable, rigid all-trans polyproline-II helical structure, which enhances the trans-amide preference through a cascade effect[1].
(2S,3aS,7aS)-octahydroindole‑2‑carboxylic acid converts into its N‑Boc‑protected methyl ester and then proceeds through high‑yield stereoselective α‑alkylation to yield enantiopure α‑substituted derivatives for peptide insertion[2].
(2S,3aS,7aS)-octahydroindole‑2‑carboxylic acid exhibits higher hydrophobicity than common hydrophobic amino acids and proline[3].
