Background
Function:Inhibitory receptor acting as a major negative regulator of T-cell responses. The affinity of CTLA4 for its natural B7 family ligands, CD80 and CD86, is considerably stronger than the affinity of their cognate stimulatory coreceptor CD28
Subcellular Localization:Cell membrane; Single-pass type I membrane protein
Expression:
Tissue_specificity:It is widely expressed, with the highest expression level in lymphoid tissues. It can be detected in activated T cells, and its expression level after activation is 30 to 50 times lower than that of the cell surface stimulatory co-receptor CD28.
Isoforms & Post-Translational Modification:P16410 has 5 isomers: P16410-1: 24656 Da (predicted); P16410-2: 6560 Da (predicted); P16410-3: 6745 Da (predicted); P16410-4: 8855 Da (predicted); P16410-5: 19145 Da (predicted).
N-glycosylation is important for dimerization;Phosphorylation at Tyr-201 prevents binding to the AP-2 adapter complex, blocks endocytosis, and leads to retention of CTLA4 on the cell surface
Subunit:Homodimer; disulfide-linked (PubMed:11279501, PubMed:11279502, PubMed:21156796, PubMed:28484017, Ref.23). Binds to CD80/B7-1 and CD86/B7.2 (PubMed:11279501, PubMed:11279502, PubMed:28484017). Interacts with ICOSLG (PubMed:28484017)

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
