  • Immunohistochemical analysis of paraffin-embedded human Testis tissue using DDX4 antibody was performed. The section was pretreated using high-temperature and high-pressure mediated EDTA antigen retrieval buffer (pH 9.0), for 5 minutes. The tissues were incubated with primary antibody (HY-P81463, 1:100 dilution) at room temperature for 60 minutes. Detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. The tissues were counterstained with hematoxylin and mounted with neutral balsam mounting medium.

  • Tyramide signaling amplification based immunofluorescence was performed on paraffin-embedded human Testis tissue using DDX4 antibody. Antigen retrieval was performed in EDTA buffer pH 9.0 (95 °C, 20 min) followed by cooling to RT. Then incubated with primary antibody (HY-P81463, 1:200 dilution) at room temperature for 60 minutes and HRP conjugated secondary antibody for 10 minutes. Fluorescence was then developed with Vari Fluor 532 TSA (200×)(HY-D1832). The tissues were counterstained with DAPI and mounted with Anti-fade mounting medium.

Background
Function:ATP-dependent RNA helicase required during spermatogenesis (PubMed:10920202, PubMed:21034600). Required to repress transposable elements and preventing their mobilization, which is essential for the germline integrity (By similarity). Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and governs the methylation and subsequent repression of transposons (By similarity). Involved in the secondary piRNAs metabolic process, the production of piRNAs in fetal male germ cells through a ping-pong amplification cycle (By similarity). Required for PIWIL2 slicing-triggered piRNA biogenesis: helicase activity enables utilization of one of the slice cleavage fragments generated by PIWIL2 and processing these pre-piRNAs into piRNAs (By similarity)
Subcellular Localization:Cytoplasm; Cytoplasm, perinuclear region
Expression:
Tissue_specificity:It is expressed only in the ovaries and testes. It is also expressed in migratory primordial germ cells in the gonadal crest region of both sexes.
Isoforms & Post-Translational Modification:Q9NQI0 has 4 isomers: Q9NQI0-1: 79308 Da (predicted); Q9NQI0-2: 75821 Da (predicted); Q9NQI0-3: 63211 Da (predicted); Q9NQI0-4: 77056 Da (predicted).
Subunit:Interacts with RANBP9 (PubMed:27622290). Interacts with RANBP10. Interacts with PIWIL2 and MAEL.

  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
