Description

(Arg)9,TAMRA-labeled acetate is a TAMRA-labeled, membrane-permeable cationic cell-penetrating polyarginine peptide. (Arg)9,TAMRA-labeled acetate acts as a cell-penetrating peptide for drug delivery[1].

In Vitro

(Arg)9,TAMRA-labeled acetate (1-5 μM; 3 h, overnight at 37 °C) shows low, dose-dependent permeation into NIH 3T3 mouse embryonic fibroblast cells, with no statistically significant difference in permeation between 37 °C (active endocytosis plus passive diffusion) and 4 °C (passive diffusion only) at 2 μM[1].
(Arg)9,TAMRA-labeled acetate (5 μM; 3 h at 37 °C) shows low permeation into NIH 3T3 mouse embryonic fibroblast cells, as indicated by sparse punctate intracellular fluorescence[1].
(Arg)9,TAMRA-labeled acetate (10 μM; 24 or 48 h at 37 °C) shows no significant cytotoxicity toward NIH 3T3 mouse embryonic fibroblast cells[1].
(Arg)9,TAMRA-labeled acetate (2 μM; 3 h at 37 °C or 4 °C) shows low permeation into H9 human embryonic stem cells, with no statistically significant difference in permeation between 37 °C and 4 °C, and a bimodal fluorescence distribution at 37 °C[1].
(Arg)9,TAMRA-labeled acetate (5 μM; 3 h at 37 °C) shows low permeation into H9 human embryonic stem cells, as indicated by sparse punctate intracellular fluorescence in cell colonies[1].
(Arg)9,TAMRA-labeled acetate (10 μM; 24 or 48 h at 37 °C) shows no significant cytotoxicity toward H9 human embryonic stem cells[1].

Cell Cytotoxicity Assay[1]

Cell Line: NIH 3T3 mouse embryonic fibroblast cells
Concentration: 10 μM
Incubation Time: 24 h; 48 h at 37 °C
Result: Caused no significant difference in NIH 3T3 cell viability relative to vehicle controls or other peptides.

Cell Cytotoxicity Assay[1]

Cell Line: H9 human embryonic stem cells
Concentration: 10 μM
Incubation Time: 24 h; 48 h at 37 °C
Result: Caused no significant difference in H9 human embryonic stem cell viability relative to vehicle controls or other peptides.
In Vivo

(Arg)9,TAMRA-labeled acetate (20 μM; in solution; overnight; room temperature) shows no membrane adsorption or internal permeation in dechorionated Drosophila melanogaster embryos/larvae, with no viability loss or larval transition observed in treated hosts[1].
(Arg)9,TAMRA-labeled acetate (20 μM; in solution; overnight; room temperature) achieves notable internal permeation in dechorionated-and-dewaxed Drosophila melanogaster embryos, with viability loss unrelated to reagent exposure and all treated hosts remaining in the embryo stage[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: Drosophila melanogaster[1]
Dosage: 20 μM

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
