Benzoyl isothiocyanate is an aromatic isothiocyanate derivative with antibacterial and acaricidal activities. Benzoyl isothiocyanate inhibits the growth of specific oral bacteria, and also serves as an intermediate for the synthesis of heterocyclic compounds such as thiazolidine and quinazoline derivatives. Benzoyl isothiocyanate exhibits acaricidal activity against Dermatophagoides farina (dust mite) and D. pteronyssinus (house dust mite). Benzoyl isothiocyanate can be used in research related to bacterial infection, pest control and organic synthesis.

Biological Activity

In Vitro
Benzoyl isothiocyanate can react with various arylamine substrates to form thiourea intermediates, which can be converted into a series of heterocyclic products including thiazolidines, oxazolidines, imidazolidinones, thiazines and tetrahydroquinazolines via desulfurization, acid cyclization and other transformations[1].
Benzoyl isothiocyanate (0.10-5.0 mg/disc; 2 days) potently inhibits the growth of Clostridioides difficile ATCC 9689, Clostridium perfringens ATCC 13124, and Escherichia coli ATCC 11775 at the dose of 5.0 mg/disc, with its activity decreasing in a dose-dependent manner: it only exerts weak inhibitory effects on the three strains mentioned above at 0.25 mg/disc; at 0.10 mg/disc, it only shows weak inhibitory effects on Clostridium strains but has no inhibitory effect on E. coli[2].
Benzoyl isothiocyanate (0.05-2.0 mg/disc; 2 days) exerts dose-dependent anaerobic growth inhibitory effects against Porphyromonas asaccharolytica ATCC 25260, Porphyromonas gingivalis ATCC 33277, Streptococcus sobrinus ATCC 27607, and Streptococcus mutans ATCC 25175[3].
Benzoyl isothiocyanate exhibits acaricidal activity against Dermatophagoides farina (house dust mite) and D. pteronyssinus (European house dust mite), with LD50 values of 13.8 and 7.63 µg/cm2 respectively[4].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: 5 mg/mL (30.64 mM); Suspended solution; Need ultrasonic
This protocol yields a suspended solution of 5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
