Biological Activity
Guide (The following is our recommended solution. This solution is merely a guideline and should be modified according to your specific needs)[2].
1. Preparation of stock solution: Dissolve the dye using high-quality DMSO and prepare a stock solution of 2-10 mM.
2. Preparation of working solution: Dilute the stock solution with preheated serum-free buffer (such as Hanks balanced salt solution (HBSS) containing 20 mM HEPES) to prepare a working solution with a final concentration of 1-20 μM.
3. Staining of adherent cells:
(1) Remove the original culture medium.
(2) Add the working solution to the cells.
(3) Incubate at 37℃ for 15 to 45 minutes.
(4) Replace with a fresh, fully grown culture medium.
(5) Before imaging, allow the cells to recover for at least 15-30 minutes to reduce the background signal.
4. Suspension cell staining: Centrifuge the cells to collect them, resuspend them in the working solution, incubate, wash, and then resuspend them in fresh culture medium.
5. Detection: The detection is carried out using a channel with excitation/emission wavelengths of 577/602 nm.
