Description

PA-SiR-Halo is a photoactivatable fluorophore targeting HaloTag. When bound to HaloTag, PA-SiR-Halo stabilizes its fluorescent form under physiological pH. After conjugation and photoactivation, it exhibits enhanced resistance to nucleophiles, and enables live-cell confocal imaging, fixed-cell single-molecule localization microscopy imaging, live-cell single-particle tracking, as well as live-cell single-molecule localization microscopy imaging of dynamic mitochondrial structures without the need for washing[1][2].

In Vitro

After conjugation with purified HaloTag protein, PA-SiR-Halo maintains stable fluorescence upon UV activation under physiological pH conditions, and its resistance to cysteamine-mediated fluorescence loss (EC50 = 3.1 mM) is higher than that of unconjugated PA-SiR[1].
PA-SiR-Halo (0.5 μM; 2 h) labels H2B-HaloTag in live U-2 OS cells. After UV activation, it reaches a signal-to-noise ratio as high as 32, with no pre-activation fluorescence signal and stable post-activation signal[1].
PA-SiR-Halo enables high-resolution fixed-cell single-molecule localization microscopy (SMLM) imaging of Cep41-HaloTag-labeled microtubules in U-2 OS cells, with a measured average microtubule diameter of 38.7 nm[1].
PA-SiR-Halo (0.5 μM; 1 h) enables live-cell single-particle tracking of β2AR-HaloTag in U-2 OS cells[1].
PA-SiR-Halo enables live-cell single-molecule localization microscopy (SMLM) imaging of dynamically changing outer mitochondrial membranes labeled with TOMM20-HaloTag in U-2 OS cells, resolving transient structures such as inter-mitochondrial tubules and fission events[1].
