Palmitoyl glutathione  (Synonyms: PGSH)

Palmitoyl glutathione (PGSH) is a palmitoylated peptide derivative and a liver-targeted liposome-forming agent. Palmitoyl glutathione can form liposome-like vesicles with cholesterol, which can encapsulate water-soluble solutes. Palmitoyl glutathione can be used in studies related to Acetaminophen (HY-66005)-induced liver necrosis^[1].

Palmitoyl glutathione (60 μmol/mL total lipids; PGSH:cholesterol molar ratio 2:1) forms stable lamellar liposome-like structures with cholesterol, achieving a [¹⁴C]sucrose encapsulation ratio of 14.9%, while no functional encapsulation occurs without cholesterol^[1].
Palmitoyl glutathione (formulated with cholesterol at 2:1 molar ratio; up to 2 h, 37 °C) liposomes exhibit stability in fresh rat plasma at 37 °C comparable to conventional phosphatidylcholine liposomes, retaining over 80% of encapsulated marker after 2 h^[1].

Palmitoyl glutathione (30 μmol total lipids per rat; i.v.; single bolus injection) administered to male Wistar rats show rapid blood clearance, high liver accumulation (67.42% of injected dose at 4 hours), low accumulation in spleen, kidneys, and lungs^[1].
Palmitoyl glutathione (81.8 mg/kg; i.v.; single bolus injection) formulated with phosphatidylcholine, administered to male ddY mice 30 minutes before acetaminophen challenge, significantly suppresses Acetaminophen-induced plasma GOT elevation with a mean absorbance of 0.496^[1].
