Photosensitizer-10 is a glutathione (GSH)-activatable photosensitizer. Description

Photosensitizer-10 is a glutathione (GSH)-activatable photosensitizer. Photosensitizer-10 shows minimal photoactivity in the absence of glutathione, and generates a highly active species upon reaction with glutathione to induce photo-cytotoxicity. Photosensitizer-10 exhibits selective photo-cytotoxicity toward cancer cells with elevated intracellular glutathione levels while sparing normal cells. Photosensitizer-10 can be used for the research of cancer, such as lung carcinoma and skin carcinoma[1].

In Vitro

Photosensitizer-10 (Compound 3) (100 μM) shows reduced absorbance at 470 nm, limiting excitation to the singlet excited state and suppressing photo-induced reactive oxygen species (ROS) formation[1].
Photosensitizer-10 (3-100 μM; 2 h) shows negligible photo-induced degradation of BSA[1].
Photosensitizer-10 (1-10 μM; 24 h) shows selective photo-cytotoxicity toward GSH-overexpressing A549 and A431 cancer cells, while sparing normal WI-38 fibroblasts[1].
Photosensitizer-10 can release the active substance Compound 2 after being activated by GSH[1].

Cell Cytotoxicity Assay[1]

Cell Line: A549 human lung carcinoma cells, A431 human squamous carcinoma cells, WI-38 human lung fibroblasts
Concentration: 1, 3, 10 μM
Incubation Time: 15 min (light irradiation); 24 h (post-irradiation incubation)
Result: Exhibited no dark toxicity across all tested cell lines.
Upon light irradiation, induced dose-dependent cytotoxicity in A549 and A431 cancer cells, while WI-38 normal fibroblasts remained largely unaffected.
At 10 μM, reduced cell viability to ~20% in A549 cells and ~15% in A431 cells, but kept viability near 100% in WI-38 cells.
