Materials
Assay buffer: 50 mM Tris, 25 mM MgCl2, 50 mM KCl, 0.5 mM DTT, pH 8.0

Acetoacetyl Coenzyme A Sodium Salt Hydrate (AACoA)
Procedure
1. Dilute Rat ACAT2 to 0.05 μg/mL in assay buffer.
2. Prepare substrate mixture containing 100 μM CoA and 30 μM AACoA in assay buffer.
3. Experimental group: 50 μL protein + 50 μL substrate.
Blank group: 50 μL assay buffer + 50 μL substrate.
4. Read in kinetic mode for 10 minutes at an absorbance of 303 nm.
5. Calculate specific activity:
