Background
Apolipoprotein A-V (APOA5) is a minor apolipoprotein primarily associated with HDL and, to a lesser extent, with VLDL, and may also be linked to chylomicrons. It plays a crucial role in regulating plasma triglyceride (TG) levels, acting as a potent stimulator of apo-CII lipoprotein lipase (LPL) TG hydrolysis and as an inhibitor of the hepatic VLDL-TG production rate, without affecting the VLDL-apoB production rate. APOA5 activates lecithin:cholesterol acyltransferase (LCAT) poorly and does not enhance cholesterol efflux from macrophages. Additionally, it exhibits binding affinity for heparin and interacts with GPIHBP1. Furthermore, APOA5 engages in an interaction with SORL1, influencing its internalization and sorting, either to lysosomes for degradation or to the trans-Golgi network.
