Background
Bcl-XL protein forms heterodimers with BAX, BAK, or BCL2, with heterodimerization with BAX not being essential for its anti-apoptotic activity. Additionally, it interacts with isoform 1 of SIVA1, inhibiting its anti-apoptotic function. The protein also engages with IKZF3 and RTL10/BOP. Furthermore, interactions with DNM1L and CLTA suggest the potential formation of a complex in synaptic vesicles that includes clathrin and MFF. Notably, Bcl-XL interacts with NLRP1, specifically via the loop between motifs BH4 and BH3, but does not engage with NLRP2, NLRP3, NLRP4, PYCARD, or MEFV. It also interacts with BECN1, indicating its involvement in autophagy regulation.
Verified Bioactivity
Measured by its binding ability in a functional ELISA. Immobilized human BID at 10 μg/mL can bind biotinylated mouse BCL2L1. The ED50 for this effect is 5.195 ng/mL.
