BRD3 Protein, Human (BD2, His) is the recombinant human-derived BRD3, expressed by E. coli, with N-6*His labeled tag.
BRD3 is a chromatin reader that recognizes and binds acetylated histones, thereby controlling gene expression and remodeling chromatin structures. It recruits transcription factors and coactivators to target gene sites and activates RNA polymerase II machinery for transcriptional elongation. In vitro, BRD3 binds acetylated lysine residues on the N-terminus of histones H2A, H2B, H3, and H4. During endoderm differentiation, BRD3 associates with the long non-coding RNA (lncRNA) DIGIT, undergoing liquid-liquid phase separation to promote binding to histone H3 acetylated at Lys-18 (H3K18ac), thereby inducing endoderm gene expression. Additionally, BRD3 binds non-histone acetylated proteins such as GATA1 and GATA2 (by similar): it regulates transcription by promoting the binding of transcription factor GATA1 to its targets.
