Background
CHMP1B is a probable peripherally associated component of the endosomal sorting required for transport complex III (ESCRT-III), which is involved in multivesicular bodies (MVBs) formation and sorting of endosomal cargo proteins into MVBs. MVBs contain intraluminal vesicles (ILVs) generated by invagination and scission from the endosomal limiting membrane, primarily delivered to lysosomes for degradation of membrane proteins (e.g., stimulated growth factor receptors, lysosomal enzymes, and lipids). The MVB pathway requires sequential functions of ESCRT-O, -I, -II, and -III complexes. ESCRT-III proteins typically dissociate from the invaginating membrane before ILV release. The ESCRT machinery also operates in topologically equivalent membrane fission events, such as cytokinesis and enveloped virus budding (e.g., HIV-1 and other lentiviruses). ESCRT-III proteins mediate vesicle extrusion and/or membrane fission, potentially in conjunction with the AAA ATPase VPS4. It participates in cytokinesis by recruiting VPS4A/VPS4B and SPAST to the midbody and facilitates HIV-1 p6- and p9-dependent virus release.
