Background

CK1d Protein, an essential serine/threonine-protein kinase, intricately regulates cellular growth and survival processes encompassing Wnt signaling, DNA repair, and circadian rhythms. Demonstrating versatility, CK1d phosphorylates a diverse array of proteins, including connexin-43/GJA1, MAP1A, SNAPIN, MAPT/TAU, TOP2A, DCK, HIF1A, EIF6, p53/TP53, DVL2, DVL3, ESR1, AIB1/NCOA3, DNMT1, PKD2, YAP1, PER1, and PER2. It plays a central role in the circadian clock, determining the period length through the regulation of PER1 and PER2 phosphorylation. Additionally, CK1d influences the WNT3A signaling pathway, neuronal outgrowth, ubiquitination of YAP1, DNA-binding activity of DNMT1, and numerous other cellular processes. This multifaceted kinase emerges as a key player in coordinating diverse cellular functions and signaling pathways.

Biological Activity

The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecencelabeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 12.5 nM, the conversion rate is about 28%.
