Background
DENND1A, a guanine nucleotide exchange factor (GEF), plays a pivotal role in orchestrating clathrin-mediated endocytosis through the activation of RAB35. Its function involves catalyzing the exchange of GDP to GTP, thereby converting the inactive GDP-bound form of RAB35 into its active GTP-bound state. Notably, DENND1A is a key regulator of clathrin-mediated endocytosis processes, influencing synaptic vesicle dynamics and facilitating the exit from early endosomes. Additionally, DENND1A exhibits binding affinity for phosphatidylinositol-phosphates (PtdInsPs), with a preference for PtdIns(3)P.
