Background

IgE, a crucial component of immunoglobulin heavy chains, plays a pivotal role in the intricate orchestration of humoral immunity. Serving as receptors during the recognition phase, membrane-bound immunoglobulins initiate clonal expansion and B lymphocyte differentiation into plasma cells upon antigen binding. In the effector phase, secreted immunoglobulins facilitate the elimination of bound antigens. The antigen-binding site, marked by remarkable affinity for specific antigens, results from the collaboration between the variable domains of one heavy chain and its associated light chain. Through V-(D)-J rearrangement and subsequent somatic hypermutations, the variable domains undergo affinity maturation, enhancing specificity for particular antigens. Structurally, each immunoglobulin consists of two identical heavy chains and two identical light chains, disulfide-linked. The N-terminal variable regions of the heavy and light chains constitute the antigen-binding sites, while the C-terminal constant regions of the heavy chains interact with immune receptors, driving effector functions. This dynamic interplay defines the versatile and precise role of IgE in orchestrating humoral immune responses.
