Background

KARS protein is responsible for catalyzing the specific attachment of an amino acid to its cognate tRNA through a two-step reaction: the amino acid is initially activated by ATP to form AA-AMP and then transferred to the acceptor end of the tRNA. In addition to its role in protein synthesis, when secreted, KARS acts as a signaling molecule, inducing immune responses by activating monocyte/macrophages. Furthermore, KARS plays a crucial role in synthesizing the signaling molecule diadenosine tetraphosphate (Ap4A), leading to the disruption of the complex between HINT1 and MITF and the subsequent activation of MITF transcriptional activity. In the context of microbial infection, KARS interacts with the HIV-1 virus GAG protein, facilitating the selective packaging of tRNA(3)(Lys), which serves as the primer for reverse transcription initiation.
