Background
HGK is a serine/threonine kinase involved in the response to environmental stress and cytokines such as TNF-alpha. It appears to act upstream of the JUN N-terminal pathway. As an activator of the Hippo signaling pathway, it plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. MAP4Ks function in parallel to and partially redundantly with STK3/MST2 and STK4/MST2 in the phosphorylation and activation of LATS1/2, establishing MAP4Ks as components of the expanded Hippo pathway. Additionally, HGK phosphorylates SMAD1 at Thr-322.
Verified Bioactivity
The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 0.15 nM, the conversion rate is about 23%.
