Background
The NCK1 protein serves as an adapter protein that associates with tyrosine-phosphorylated growth factor receptors such as KDR and PDGFRB, as well as their cellular substrates. It plays a pivotal role in maintaining low levels of EIF2S1 phosphorylation by promoting its dephosphorylation through PP1. While not directly involved in detecting DNA damage by ATM/ATR, NCK1 is crucial for the efficient activation of downstream effectors, including CHEK2, in the DNA damage response. Additionally, NCK1 contributes to ELK1-dependent transcriptional activation in response to activated Ras signaling and modulates EIF2AK2/PKR activation by dsRNA. Its involvement in cell adhesion and migration is suggested through interactions with ephrin receptors.
