NCK1 Protein, Human (His)

NCK1 protein is an adapter protein that binds to tyrosine phosphorylated receptors (KDR, PDGFRB) and cellular substrates and plays a key role in the dephosphorylation of EIF2S1 by PP1. It contributes to the efficient activation of DNA damage response effector (CHEK2) and ELK1-dependent transcriptional activation in Ras signaling. NCK1 Protein, Human (His) is the recombinant human-derived NCK1 protein, expressed by E. coli , with N-6*His labeled tag.

Description

NCK1 protein is an adapter protein that binds to tyrosine phosphorylated receptors (KDR, PDGFRB) and cellular substrates and plays a key role in the dephosphorylation of EIF2S1 by PP1. It contributes to the efficient activation of DNA damage response effector (CHEK2) and ELK1-dependent transcriptional activation in Ras signaling. NCK1 Protein, Human (His) is the recombinant human-derived NCK1 protein, expressed by E. coli , with N-6*His labeled tag.

Background

The NCK1 protein serves as an adapter protein that associates with tyrosine-phosphorylated growth factor receptors such as KDR and PDGFRB, as well as their cellular substrates. It plays a pivotal role in maintaining low levels of EIF2S1 phosphorylation by promoting its dephosphorylation through PP1. While not directly involved in detecting DNA damage by ATM/ATR, NCK1 is crucial for the efficient activation of downstream effectors, including CHEK2, in the DNA damage response. Additionally, NCK1 contributes to ELK1-dependent transcriptional activation in response to activated Ras signaling and modulates EIF2AK2/PKR activation by dsRNA. Its involvement in cell adhesion and migration is suggested through interactions with ephrin receptors. NCK1 engages in a diverse array of protein interactions, including those with EGFR, PAK1, SOS1, BLNK, PLCG1, VAV1, SOCS7, RALGPS1, CAV2, ADAM15, FASLG, RASA1, MINK1, FLT1, KDR, EPHB1, EPHA2, and PDGFRB, highlighting its intricate role in orchestrating cellular signaling pathways and molecular complexes.
