ALIX is a multifunctional protein involved in endocytosis, multivesicular body (MVB) biogenesis, membrane repair, cytokinesis, apoptosis, and tight junction maintenance. As a Class E VPS protein, it facilitates cargo sorting into intralumenal vesicles (ILVs) within MVBs by interacting with ESCRT complexes (ESCRT-O, -I, -II, -III) and binding lysobisphosphatidic acid (LBPA). It acts as an adapter for ESCRT-III proteins (e.g., CHMP4) and is essential for cytokinesis completion. ALIX regulates exosome biogenesis with SDC1/4 and SDCBP and may modulate apoptosis and cell proliferation. By interacting with F-actin, PARD3, and TJP1, it ensures proper actomyosin-tight junction assembly in epithelial polarity (By similarity). Additionally, ALIX supports HIV-1 budding by functionally replacing TSG101 via CHMP4B interaction, highlighting its role in viral release and topologically similar membrane fission events.
