The PRAK/MAPKAPK5 Protein, belonging to the serine/threonine kinase family, functions as a tumor suppressor and responds to cellular stress and proinflammatory cytokines by getting activated through phosphorylation by MAP kinases, including MAPK1/ERK, MAPK14/p38-alpha, and MAPK11/p38-beta. While initially located in the nucleus, upon phosphorylation and activation, it translocates to the cytoplasm. A notable target of this kinase is the heat shock protein HSP27, phosphorylating it at physiologically relevant sites. This gene displays two alternatively spliced transcript variants encoding distinct isoforms.
