Background
Band 3 functions as both an electroneutral anion exchanger across the cell membrane and a structural protein. As part of the ankyrin-1 complex in the erythrocyte membrane, it maintains membrane flexibility, stability, and cell shape through interactions with cytoskeletal proteins, glycolytic enzymes, and hemoglobin. It mediates 1:1 inorganic anion exchange in erythrocytes and chloride-bicarbonate exchange in the kidney, essential for urine acidification. During microbial infection, Band 3 acts as a receptor for Plasmodium falciparum (isolate 3D7) MSP9 and MSP1, facilitating merozoite invasion of erythrocytes.
