Biological Activity
UBE3A Protein, Human is the recombinant human-derived UBE3A, expressed by E. coli.
UBE3A is an E3 ubiquitin-protein ligase that accepts ubiquitin from an E2 ubiquitin-conjugating enzyme as a thioester and transfers it to substrates. Identified substrates include BMAL1, ARC, LAMTOR1, RAD23A, RAD23B, MCM7 (involved in DNA replication), annexin A1, the PML tumor suppressor, and the cell cycle regulator CDKN1B. Additionally, UBE3A may function as a cellular quality control ubiquitin ligase by facilitating the degradation of cytoplasmic misfolded proteins and promotes its own degradation in vivo. It plays a critical role in circadian clock regulation by ubiquitinating the core clock component BMAL1, leading to its proteasomal degradation. UBE3A acts as a transcriptional coactivator of progesterone receptor PGR upon progesterone activation. By similarity, it regulates synaptic development by mediating ubiquitination and degradation of ARC and controls synaptic remodeling in neurons by ubiquitinating and degrading LAMTOR1, thereby limiting mTORC1 signaling and activity-dependent synaptic remodeling. UBE3A also synergizes with WBP2 to enhance PGR activity.
