5'-ODMT cEt N-Bz A Phosphoramidite (Amidite) is a nucleoside phosphoramidite monomer used to synthesize locked nucleic acid (LNA) analog oligonucleotides. It can be used as a building block of antisense oligonucleotides (ASOs) to target complementary RNA sequences. 5'-ODMT cEt N-Bz A Phosphoramidite (Amidite) locks the furanose ring into an N-type conformation through 2',4'-constrained ethyl (cEt) modification, enhancing hybridization affinity and mismatch discrimination with RNA, while significantly improving the resistance of oligonucleotides to exonuclease digestion. 5'-ODMT cEt N-Bz A Phosphoramidite (Amidite) mediates RNase H-dependent mRNA degradation or inhibits translation by forming a stable hybrid with RNA, thereby achieving gene expression regulation. 5'-ODMT cEt N-Bz A Phosphoramidite (Amidite) is mainly used in the development of antisense drugs, gene function research and oligonucleotide synthesis related to disease treatment.

5'-ODMT cEt N-Bz A Phosphoramidite (Amidite) is used in oligonucleotide synthesis experiments to synthesize locked nucleic acid (LNA) analog oligonucleotides with high affinity and exonuclease resistance, which can effectively hybridize with complementary RNA sequences and regulate gene expression[1].
LNA has a wide range of applications in molecular biology, genetic research and disease treatment[1]
1. In molecular biology techniques, its nucleoside triphosphates can be used as substrates for primer extension and PCR experiments: 9°NTM DNA polymerase performs best in PCR amplification containing LNA nucleotides, and LNA-modified probes and primers can improve the melting temperature (Tm value), stability and mismatch discrimination ability of real-time PCR (qPCR), which is suitable for SNP detection, pathogen identification and multiplex analysis.
2. In terms of gene expression regulation, LNA can be used as an antisense oligonucleotide (ASO) to inhibit gene expression. LNA can mediate RNase H-dependent mRNA degradation by forming stable hybrids and is used for small RNA research and gene silencing.
3. In disease diagnosis and treatment, the high affinity and nuclease resistance of LNA make it suitable for complex samples such as FFPE tissues and biological fluids. LNA can also be used for antisense drug development and is an important tool for gene therapy. It also improves detection sensitivity and specificity in techniques such as in situ hybridization (FISH) and microarray analysis.
