    Mucin 6 (MUC6) is a secreted gel-forming mucin that constitutes a major component of gastric mucus and is predominantly expressed in the deep gastric glands, particularly the pyloric and mucous neck gland compartments[1][2][3]. MUC6 contributes to the organization of the gastric mucus barrier together with MUC5AC, forming a protective mucosal system that supports resistance to gastric acid, pepsin, and microbial challenge[4][5][6]. Mechanistically, the heavily glycosylated structure of MUC6 participates in the formation of a viscoelastic mucus layer, and its carbohydrate side chains interact with bicarbonate and other protective factors that help maintain mucosal integrity[4][5]. In gastric physiology, MUC6 exhibits a distinct expression pattern compared with the related gastric mucin MUC5AC, which is primarily localized to the superficial foveolar epithelium, whereas MUC6 is concentrated within glandular regions[1][2][3][7]. This spatial specialization makes MUC6 a useful marker for defining gastric epithelial differentiation and gastric-type mucin phenotypes in experimental pathology studies[7]. Altered MUC6 expression has been reported during gastric carcinogenesis and other gastric pathological conditions, supporting its utility as a biomarker in studies of disease progression and mucosal transformation[4][8][9]. Experimental evidence further indicates that enhancement of MUC6 expression accompanies cytoprotective responses in gastric injury models, where increased MUC6 production is associated with reinforcement of the mucus barrier[10].

    Tissue_specificity:It is expressed in the regeneration zone of the gastric antrum, the mucosa of the gastric body, and the mucosa of the gastric notch. It is also expressed in the deep mucous glands of the gastric antrum. It is overexpressed in gastric epithelium infected with Helicobacter pylori. It is highly expressed in the Brenner's glands of the duodenum, gallbladder, seminal vesicle, central cells and ducts of the pancreatic acinar, and glands surrounding the common bile duct.
