Immunohistochemical analysis of paraffin-embedded rat placenta tissue using Vinculin Antibody (HY-P85789, 1/800). The section was pretreated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked with quick block buffer for 0.5 hours at room temperature, washed with PBS and PBST, and then incubated with the primary antibody overnight at 4℃. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

Function
Vinculin is an Actin filament (F-actin)-binding protein involved in cell-matrix adhesion and cell-cell adhesion. Regulates cell-surface E-cadherin expression and potentiates mechanosensing by the E-cadherin complex. May also play important roles in cell morphology and locomotion

Isoforms & Post-Translational Modification
P18206 has 3 isomers: P18206-1: 123799 Da (predicted); P18206-2: 116722 Da (predicted); P18206-3: 24904 Da (predicted).
Phosphorylated; on serines, threonines and tyrosines. Phosphorylation on Tyr-1133 in activated platelets affects head-tail interactions and cell spreading but has no effect on actin binding nor on localization to focal adhesion plaques (By similarity);Acetylated; mainly by myristic acid but also by a small amount of palmitic acid -
