Kemptide-FITC
Kemptide-FITC is a fluorescently labeled Kemptide (HY-P0248). Kemptide is a synthetic heptapeptide that acts as a specific substrate for cAMP-dependent protein kinase (PKA). Kemptide-FITC enables the quantification of PKA activity and can also indirectly determine cAMP concentrations through the activation of PKA in sperm insoluble fractions. Kemptide-FITC is applicable for kinase mobility shift assay (KiMSA), and its fluorescent signal shows a linear response with increasing peptide amount within a specific range on agarose gels.

Biological Activity
Kemptide-FITC serves as a non-radioactive substrate for KiMSA, enabling quantitative detection of the PKA activity of recombinant PKA-C. Its phosphorylated and non-phosphorylated forms can be clearly separated via agarose gel electrophoresis, with no spectral drift interfering with fluorescence quantitative analysis[1].
Kemptide-FITC (non-phosphorylated) exhibits a linear fluorescence signal response in the range of 50 to 300 pmoles when separated by agarose gel electrophoresis[2].
Kemptide-FITC (30 μM; 25 min at 37°C) serves as a substrate for the recombinant catalytic subunit of protein kinase A (PKA) α. Detection via KiMSA reveals a linear relationship between the enzyme concentration (53-533 ng/mL) and the phosphorylation level of Kemptide-FITC[2].
