Description

NHS-activated 2-octynoic acid-BSA is an immunogen used to establish a mouse model of primary biliary cholangitis (PBC). When used in combination with Complete Freund's Adjuvant (HY-153808), NHS-activated 2-octynoic acid-BSA stimulates the body's immune system, which recognizes it as a foreign antigen and triggers an immune response against autologous biliary epithelial cells. In Vitro

2-octynoic acid (20 min (0°C stirring); overnight (ambient temp warming); 3 h (BSA coupling)) is successfully conjugated to BSA via lysine residues following a multi-step chemical synthesis and purification workflow[1].

In Vivo

2-octynoic acid-BSA (100 μg per mouse; i.p.; initial in Complete Freund’s Adjuvant, boosts every 2 weeks in Incomplete Freund’s Adjuvant) induces PBC-like autoimmune cholangitis in female C57BL/6J mice, including 100% seroconversion to anti-PDC-E2 antibodies by 8 weeks, elevated Th1 cytokines, liver lymphoid infiltration, and bile duct lesions consistent with human PBC[1].

Decreased liver CD4/CD8 ratio to 2.03 at 12 weeks post-immunization compared to controls.
Increased total liver lymphoid cell number to at 12 weeks post-immunization.
Increased absolute numbers of liver CD8+ T cells and CD19+ B cells at 12 weeks post-immunization.
Increased frequency of liver CD8+ T cells co-expressing CD44 at 12 weeks post-immunization.
