NS2B/NS3-IN-3 hydrochloride is an inhibitor of Flavivirus NS2B-NS3 protease, with an IC50 of 0.32 μM against Zika virus NS2B-NS3 protease. NS2B/NS3-IN-3 hydrochloride also exhibits inhibitory effects on dengue virus type 2 NS2B-NS3 protease and West Nile virus NS2B-NS3 protease. NS2B/NS3-IN-3 hydrochloride inhibits the intracellular replication of Zika virus. NS2B/NS3-IN-3 hydrochloride can be used in studies related to flavivirus infections.

Biological Activity
NS2B/NS3-IN-3 (compound 66) hydrochloride potently inhibits recombinant Zika virus NS2B-NS3 protease with an IC50 of 0.32 μM; it also inhibits recombinant dengue virus type 2 NS2B-NS3 protease with an IC50 of approximately 1-2 μM, and exhibits a non-competitive inhibition mode[1].
NS2B/NS3-IN-3 (1-10 μM; 48 h) hydrochloride exhibits dose-dependent antiviral activity against Zika virus (strain FLR) in human U87 glioma cells, with an EC68 of 1 μM, and shows no toxicity to U87 cells at a concentration of 10 μM[1].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of ≥ 1 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (10.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
