AMIGO2 Protein, Human (HEK293, His) is an 58-85 kDa AMIGO2 protein fused His-tag expressed in HEK 293 cells. AMIGO2 Protein, Human (HEK293, His) interacts with PDK1 to activate the Akt pathway localizated to the plasma membrane[1].
Background
AMIGO2 is a scaffold protein that regulates PDK1 membrane localization and Akt activation. This protein contains six leucine-rich repeats (LRRs), a single IgG-like domain, a transmembrane domain, and a cytosolic domain. AMIGO2 plays its role by controling PDK1 localization to the plasma membrane and then directly interacts with PDK1 to activate the Akt pathway[1]. Loss of AMIGO2 in endothelial cells (ECs) leads to apoptosis and inhibition of angiogenesis with Akt inactivation[1]. AMIGO2 is important in regulating T-cell functions, Amigo2-overexpression dampens NF-kB transcriptional activity, while Amigo2-deficiency enhances Akt but suppresses GSK-3β phosphorylation and promotes nuclear translocations of NF-kB and NFAT1 in Th-cells[1].
