Description

Bmal1 Protein, Mouse (Flag) is the recombinant mouse-derived Bmal1 protein, expressed by E. coli, with C-Flag tag.

Background

Bmal1 is a transcriptional activator that forms a core component of the circadian clock, an internal time-keeping system that generates approximately 24-hour rhythms in gene expression, metabolism, and behavior. Derived from the Latin roots 'circa' (about) and 'diem' (day), it regulates diverse physiological processes including metabolism, sleep, and cardiovascular function. The clock consists of a central pacemaker in the brain's suprachiasmatic nucleus (SCN) and peripheral oscillators in tissues, both synchronized by environmental cues like light. Bmal1 heterodimerizes with CLOCK or NPAS2 to activate transcription of E-box (5'-CACGTG-3')-containing clock-controlled genes, while PER/CRY proteins form the negative limb of the feedback loop. It also modulates pancreatic β-cell function via antioxidant genes (e.g., NFE2L2), inhibits mTORC1 signaling, and influences monocyte rhythms, neurogenesis, and spermatogenesis. The CLOCK-BMAL1 heterodimer preferentially binds 5'-CACGTGA-3' (By similarity) and noncanonical E-boxes (By similarity). Recent studies highlight its role in sepsis resistance by suppressing CD274 (PubMed:29996098) and regulating muscle metabolism (PubMed:30096135).
