Ferritin light chain/FTL Protein, Human (His)

Cat. No.: HY-P70245
COA Handling Instructions

Ferritin light chain/FTL protein is pivotal in iron homeostasis, storing iron in a soluble, non-toxic form and delivering it to cells. In kidney development, FTL mediates iron uptake in capsule cells, crucial for organogenesis. Structurally, FTL, an oligomer with light and heavy subunits forming a spherical shell, facilitates iron entry through pores. Mutations in the C-terminus disrupt pore formation, impacting iron accumulation. Ferritin light chain/FTL Protein, Human (His) is the recombinant human-derived Ferritin light chain/FTL protein, expressed by E. coli , with N-6*His labeled tag. The total length of Ferritin light chain/FTL Protein, Human (His) is 175 a.a., with molecular weight of 20-25 kDa.

Background

Ferritin light chain/FTL protein plays a pivotal role in iron homeostasis by storing iron in a soluble, non-toxic, and readily available form. The ferrous iron is taken up and subsequently deposited as ferric hydroxides after oxidation. Beyond its iron storage function, FTL is actively involved in delivering iron to cells, underscoring its significance in cellular processes. In the context of kidney development, FTL participates in mediating iron uptake in capsule cells, highlighting its role in organogenesis. Structurally, FTL exists as an oligomer comprising 24 subunits, with two types of subunits, light (L) and heavy (H), forming the spherical shell. The functional molecule features a central cavity where the insoluble mineral iron core is deposited. This intricate structure allows iron entry through pores formed between subunits. Mutations affecting the C-terminus can interfere with normal pore formation and disrupt iron accumulation.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
