FYN Protein, Human (Biotinylated, sf9, His, Flag, Avi) is the recombinant human-derived FYN, expressed by Sf9 insect cells , with Avi, His, Flag labeled tag.
Background
Fyn is a non-receptor tyrosine-protein kinase involved in regulating diverse biological processes such as cell growth/survival, adhesion, integrin-mediated signaling, cytoskeletal remodeling, motility, immune response, and axon guidance. Inactive Fyn is phosphorylated at its C-terminal tail; upon PKA activation, it associates with PTK2/FAK1 to facilitate FAK1 phosphorylation and focal adhesion targeting. It modulates adhesion/motility via CTNNB1 (β-catenin) and CTNND1 (δ-catenin) phosphorylation, and drives cytoskeletal remodeling by phosphorylating WAS, MAP2, and MAPT. Fyn promotes cell survival by inhibiting AGAP2/PIKE-A cleavage and maintains glomerular slit diaphragm integrity via NPHS1/KIRREL1/TRPC6 phosphorylation. In neural contexts, it phosphorylates DPYSL2, ARHGAP32, and SNCA. Fyn also mediates TCR-induced T-cell proliferation/differentiation by phosphorylating PTK2B/PYK2 and negatively regulates TCR signaling through PAG1-CSK interactions. Similar roles include: DAB1 phosphorylation in reelin signaling (by similar); CLNK phosphorylation in IgE receptor-activated mast cells (by similar); and CD244-dependent NK cell activation.
Verified Bioactivity
The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecence-labeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 0.0625 nM, the conversion rate is about 20%.
