FYN

FYN is a ubiquitously expressed Src-family tyrosine kinase bearing the canonical myristoylated SH3-SH2-kinase architecture, with brain-enriched and epithelial isoforms produced by alternative splicing. In T lymphocytes, FYN phosphorylates T-cell receptor chains including the zeta subunit and collaborates with LCK in antigen receptor signaling. In the central nervous system, FYN drives oligodendrocyte differentiation and myelination, organizes postsynaptic densities through scaffolds such as PSD-95, modulates NMDA-type glutamate receptors, and phosphorylates tau, connecting it to Alzheimer disease; the FYN inhibitor saracatinib entered Alzheimer clinical trials. FYN additionally participates in platelet activation and focal adhesion signaling, illustrating its broad reach across excitable and immune tissues.
