S100A4 protein, a calcium-binding molecule, plays a versatile role in numerous cellular processes, including motility, angiogenesis, cell differentiation, apoptosis, and autophagy. Functionally, it enhances cell motility and invasiveness by interacting with non-muscle myosin heavy chain IIA/MYH9, promoting filament depolymerization and increasing the amount of soluble myosin-IIA to facilitate chemotaxis. Additionally, S100A4 modulates the pro-apoptotic function of TP53 by binding to its C-terminal transactivation domain within the nucleus, leading to a reduction in TP53 protein levels. In the extracellular space, it stimulates cytokine production and acts as a chemoattractant complex with PGLYRP1 to promote lymphocyte migration via CCR5 and CXCR3 receptors.
