Background

The SERPINE1 protein is a single-chain glycoprotein composed of 379 amino acids that are mainly synthesized and secreted by platelets, megakaryocytes, hepatocytes, adipocytes, smooth muscle cells, and vascular endothelial cells. The active site of the protein is Arg346-Met347. The half-life of activated SERPINE1 at 37°C is 1–2 h, transforming spontaneously into the latent form. in the fibrinolytic system, SERPINE1 plays a physiological role as an inhibitor of fibrinogen activators. SerpinE1 drives a cell-autonomous pathogenic signaling in Hutchinson-Gilford progeria syndrome[1].
SERPINE1 was identified as a regulator of glioblastoma cell dispersal and downregulation of SERPINE1 limited the proliferation and invasion of glioma cells[3].

Biological Activity

Measured by its ability to inhibit uPA cleavage of a peptide substrate, N-carbobenzyloxy-Gly-Gly-Arg-7-amido-4-methylcoumarin (Z-GGR-AMC). The IC50 value is < 80 nM.
