SG-55 is a selective, noncompetitive and orally active AKR1C3 inhibitor with an IC50 of 5 nM and a Ki of 10 nM. SG-55 shows >2000-fold selectivity for AKR1C3 over AKR1C1, AKR1C2, and AKR1C4 (> 10 μM). SG-55 increases the ratio of reduced/oxidized nicotinamide adenine dinucleotide phosphate (NADPH/NADP +), decreases the ratio of reduced/oxidized glutathione (GSH/GSSG), and induces DNA double-strand breaks. SG-55 can overcome Osimertinib (HY-15772) resistance mediated by EGFR C797S triple mutation in non-small cell lung cancer (NSCLC) .

Synonyms: AZD-9291 (GMP); Mereletinib (GMP)
Osimertinib (AZD-9291; Mereletinib) GMP is the GMP level of Osimertinib (HY-15772). GMP small molecules works appropriately as an auxiliary reagent for cell research manufacture. Osimertinib GMP is an inhibitor that selectively targets EGFR T790M and activated EGFR mutants. Osimertinib GMP exerts multiple anti-tumor mechanisms, including radiosensitization, induction of apoptosis and ferroptosis, as well as inhibition of cancer stemness. Osimertinib GMP suppresses EGFR nuclear translocation and downstream survival signaling pathways, significantly reduces cell viability and lipid droplet content, and also exerts synergistic effects with radiotherapy or specific targeted agents to effectively overcome drug resistance and radioresistance. Formulations of Osimertinib GMP modified with liposomes or iRGD enable sustained release, tumor-specific accumulation, and breakthrough of the blood-brain barrier limitation. Osimertinib GMP can be used in research related to radioresistant glioblastoma and non-small cell lung cancer .

ER ligand-14 is an estrogen receptor (ER) ligand. ER ligand-14 can serve as a ligand for target protein for PROTAC, and can be used to develop and design degradative agents for PROTAC ERβ, such as ERB-2 (HY-180850). ERB-2 displays favorable antiproliferative activity against Osimertinib (HY-15772) resistance NSCLC cells .

ERB-2 is a selective ERβ PROTAC degrader. ERB-2 removes the inhibitory effect of ERβ on ROS, leading to the accumulation of ROS, mitochondrial damage, and ultimately triggering cell apoptosis (apoptosis). ERB-2 significantly inhibits tumor growth in the nude mouse model of NCI-H1975OR tumors. ERB-2 can be used for the study of non-small cell lung cancer .

DY3002 is a selective and highly potent EGFR inhibitor with activity in overcoming T790M-mediated drug resistance in non-small cell lung cancer. DY3002 exhibited superior inhibitory effects against EGFR T790M mutants in kinase assays (IC50 = 0.71 nM), compared to weaker inhibitory effects against wild-type EGFR (IC50 = 448.7 nM). DY3002 was significantly superior to rociletinib and osimertinib in selectivity, showing an extremely high selectivity index (SI = 632.0). In cell experiments, DY3002 had an IC50 value of 0.037 μM against H1975 cells, showing enhanced inhibitory potency. In addition, DY3002 was superior to other alternative compounds in terms of biological properties and did not cause hyperglycemia .

TopBP1-IN-4 is a TopBP1 inhibitor, with an IC50 value of 2.47 to 3.8 nM against the TopBP1 BRCT7/8 domain. TopBP1-IN-4 selectively disrupts BRCT7/8-dependent oncogenic protein-protein interactions, does not interfere with other BRCT domain-mediated functions of TopBP1, and has no effect on DNA replication. TopBP1-IN-4 restores E2F1-mediated Apoptosis in cells. TopBP1-IN-4 induces mitotic catastrophe in cells. TopBP1-IN-4 overcomes Osimertinib (HY-15772) resistance in EGFR-mutant non-small cell lung cancer models. TopBP1-IN-4 can be used in studies related to triple-negative breast cancer, ovarian cancer, non-small cell lung cancer, and acute myeloid leukemia .
