Suc-Phe-Ala-Ala-Phe-pNA (Suc-FAAF-pNA) is a chromogenic polypeptide substrate that can be used for the quantitative activity assay of intracellular subtilisin (ISP) and neutral metalloprotease (NPS) derived from Saccharomonospora canescens. Suc-Phe-Ala-Ala-Phe-pNA is hydrolyzed by the catalytic triad of mature proteases to release the chromophore pNA. Suc-Phe-Ala-Ala-Phe-pNA is applicable to enzymological studies.

Biological Activity
Suc-Phe-Ala-Ala-Phe-pNA (50 μM; 5 h) is a substrate for processed Bacillus clausii NΔ18-ISP, which exhibits proteolytic activity toward it; Suc-Phe-Ala-Ala-Phe-pNA (4-75 μM) also enables quantification of N-terminal extension peptide inhibition of NΔ18-ISP with a Ki of 1 μM[1].
Suc-Phe-Ala-Ala-Phe-pNA is hydrolyzed by the purified neutral metalloproteinase NPS from Saccharomonospora canescens sp. novus, strain 5 with a Km of 14.5 μM, kcat of 1.20 s-1[2].
