Tetrahydroaminopterin (NSC 524120) is a tetrahydropteridine analog and the tetrahydro-reduced derivative of aminopterin. Tetrahydroaminopterin is used for studies on phenylalanine hydroxylase cofactor binding and the structure-activity relationship of tetrahydrobiopterin analogs.

Tetrahydroaminopterin (NSC 524120) (up to 1 mM) shows no detectable inhibitory effect on partially purified rat liver phenylalanine hydroxylase in both spectrophotometric assays and tyrosine-forming activity assays[1].
Tetrahydroaminopterin (up to 1 mM; 10 min; 27°C) exhibits no intrinsic cofactor activity in the rat liver phenylalanine hydroxylase-catalyzed conversion of phenylalanine to tyrosine[1].
