2-Deoxy-2-fluoro-L-fucose (GMP)
2-Deoxy-2-fluoro-L-fucose (2-Fluorofucose) GMP is a fucosyltransferase inhibitor that inhibits core fucosylation modification in cells. 2-Deoxy-2-fluoro-L-fucose GMP promotes the production of defucosylated IgG1 antibodies in IgG1-expressing cell culture systems; it regulates the levels of inflammatory cytokines such as IL-1β, IL-6 and TGF-β in human induced pluripotent stem cell-derived microglia. 2-Deoxy-2-fluoro-L-fucose GMP can be used in the research of related diseases including dengue fever and Alzheimer's disease.

Description
In Vitro
2-Deoxy-2-fluoro-L-fucose GMP (250 μM; 96 h) treatment of Expi293F cells producing anti-DENV2 E protein IgG1 (C10), anti-CD4 IgG1 (YTS191), or Rituximab (HY-P9913) IgG1 generates afucosylated IgG1s that bind human FcγRIIIA with high affinity (Kd values as low as 39 nM)[1].

In Vivo
2-Deoxy-2-fluoro-L-fucose GMP (250 µM; cell culture supplementation during antibody expression) induces production of afucosylated anti-DENV2 E protein IgG1 C10, which drives severe dengue infection in Ifnar1-/-/FcγR humanized mice, matching the pathogenicity of monofucosylated C10[1].
