  3. Anti-Nicastrin Antibody (A5226A)

Anti-Nicastrin Antibody (A5226A) is a monoclonal antibody against Nicastrin and an inhibitor of γ-secretase. Anti-Nicastrin Antibody (A5226A) recognizes the fully glycosylated mature presenilin enhancer in the active γ-secretase complex and inhibits its activity via competition for substrate binding. Anti-Nicastrin Antibody (A5226A) abrogates the growth of cancer cells dependent on γ-secretase activity. Anti-Nicastrin Antibody (A5226A) serves as an imaging tool to visualize the endocytic trafficking of active γ-secretase, and also acts as a detection reagent to evaluate the endocytic efficiency of γ-secretase. Anti-Nicastrin Antibody (A5226A) can be used in studies related to non-small cell lung cancer, T-cell acute lymphoblastic leukemia and Alzheimer's disease.

Species Reactivity

Human

In Vitro

Anti-Nicastrin Antibody (A5226A) binds to human NCT at epitope R391, and interacts with both mature (active γ-secretase complex-associated) and immature forms of human NCT in NKO cells overexpressing human NCT[1].
Anti-Nicastrin Antibody (A5226A) specifically recognizes active γ-secretase complexes containing mature NCT localized in CTB-positive lipid raft microdomains on the plasma membrane of HeLa cells and NKO cells expressing human NCT[1].
Anti-Nicastrin Antibody (A5226A) dose-dependently inhibits γ-secretase activity in HEK293 cells stably expressing amyloid precursor protein- or Notch-derived γ-secretase substrates[1].
Anti-Nicastrin Antibody (A5226A) inhibits γ-secretase activity in a cell-free in vitro assay by interfering with the interaction between NCT and Notch-based substrate N100[1].
Anti-Nicastrin Antibody (A5226A) inhibits γ-secretase-dependent proliferation of A549 non-small cell lung cancer cells and DND-41 T-cell acute lymphoblastic leukemia cells, but does not affect HeLa cell viability[1].
Anti-Nicastrin Antibody (A5226A) inhibits γ-secretase activity via substrate competition in cell-free assays and causes a moderate decrease in ICN1 in DND41 T-ALL cells in vitro[3].

In Vivo

Anti-Nicastrin Antibody (A5226A) (50 mg/kg; i.p.; every 4-5 days; at least 30 days) inhibits γ-secretase activity-dependent cancer cell growth in vivo, achieving a 55.2% reduction in tumor volume at day 38 in a DND-41 xenograft model[1].
Anti-Nicastrin Antibody (A5226A) produces a significant reduction in tumor volume in human DND41 T-ALL xenotransplant models[3].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: C.B-17/Icr scid/scid Jcl (6-8 weeks old female; severe combined immunodeficiency; inoculated subcutaneously with 3×107 DND-41 cells carrying activating NOTCH1 mutations)[1]
Dosage: 50 mg/kg
Administration: i.p.; every 4-5 days; at least 30 days
Result: Reduced subcutaneous tumor volume by 55.2% at day 38 compared to control groups.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
