Function
CD328 mediates sialic acid-dependent cell binding, preferentially binding to α-2,3- and α-2,6-linked sialic acids. CD328 also binds to disialiac gangliosides (disialiac galactosylglobulin, disialiac lactotetrasaccharide ceramide, and disialiac GalNAc lactotetrasaccharide ceramide). The sialic acid recognition site may be masked by cis-interactions with sialic acids on the same cell surface. In immune responses, after ligand-induced tyrosine phosphorylation, this molecule may act as an inhibitory receptor, recruiting cytoplasmic phosphatases through its SH2 domain, thereby blocking the dephosphorylation of signaling molecules and inhibiting signal transduction. It mediates the inhibition of natural killer cytotoxicity. CD328 can also inhibit the differentiation of CD34+ cell precursors into myeloid mononuclear cell lineages and the proliferation of myeloid leukemia cells.
