Function
S100A9 is a calcium-binding S100 family protein released mainly by neutrophils and monocytes, and it commonly functions with S100A8 as the S100A8/A9 calprotectin heterodimer in innate immune inflammation[1]. Mechanistically, extracellular S100A9 engages TLR4 with CD14 as a co-receptor, inducing NF-κB activation and a pro-inflammatory cytokine response in monocytes[2]. The S100A8/A9 complex also activates inflammatory signaling through TLR4, RAGE, NF-κB, and MAPK pathways, linking leukocyte recruitment, cytokine secretion, infection defense, and inflammation-associated disease models[1][3]. In tumor models, S100A9-TLR4 signaling promoted tumor growth, whereas S100A9 deficiency, TLR4 deficiency, or ABR-215050/tasquinimod reduced EL4 lymphoma growth[4]. Compared with related isoforms, S100A9 differs from S100A8 and S100A12 because its best-characterized inflammatory role often depends on heterodimerization with S100A8 and receptor-specific TLR4/CD14 signaling[1][2][3]. For experimental applications, paquinimod reduced synovial activation, osteophyte formation, and cartilage damage in collagenase-induced osteoarthritis with high synovial activation[5].- S100A9 supports inflammation studies through TLR4/CD14-dependent cytokine induction in monocyte models[2]. - S100A9 inhibitors enable disease-model testing in tumor growth and osteoarthritis pathology[4][5].

Expression
Tissue_specificity:Calprotectin (S100A8/9) is primarily expressed in myeloid cells. Except in inflammatory states, its expression is limited to specific stages of myeloid differentiation, as both proteins are expressed in circulating neutrophils and monocytes, but absent in normal tissue macrophages and lymphocytes. Calprotectin is also expressed in the epidermis in cH2O2ic inflammatory states, such as psoriasis and malignancies. Calprotectin concentrations are higher in serum at sites of inflammation or in patients with inflammatory diseases such as rheumatoid arthritis, cystic fibrosis, inflammatory bowel disease, Crohn's disease, giant cell arteritis, Sjgren's syndrome, systemic lupus erythematosus, and progressive systemic sclerosis. Calprotectin is involved in the formation and deposition of amyloid proteins (called amyloid inclusion bodies) in the aging prostate. It is significantly upregulated in a variety of tumors, including gastric cancer, esophageal cancer, colon cancer, pancreatic cancer, bladder cancer, ovarian cancer, thyroid cancer, breast cancer, and skin cancer. -
