Carboxylesterase 2-IN-1
Carboxylesterase 2-IN-1 is a potent carboxylesterase CES2 inhibitor with an IC50 of 0.13 μM. Carboxylesterase 2-IN-1 blocks the hydrolysis of fluorescein diacetate by CES2 via non-competitive inhibition. Carboxylesterase 2-IN-1 also effectively inhibits adipogenesis induced in mouse preadipocytes.

CES2 |
Carboxylesterase 2-IN-1 (compound 26) (10-40 μM; 144 h for adipogenesis inhibition, 48 h for cytotoxicity testing) is non-toxic to 3T3-L1, Caco-2, and HepG2 cells at concentrations up to 40 μM, and potently inhibits adipogenesis and lipid droplet formation in differentiated 3T3-L1 preadipocytes, with near-complete inhibition at 40 μM[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:mouse 3T3-L1 preadipocytes, Caco-2 cells, HepG2 cells
-
Concentration:10-40 μM (adipogenesis inhibition); 0-100 μM (cytotoxicity testing)
-
Incubation Time:144 h (total adipogenesis inhibition); 48 h (cytotoxicity testing)
-
Result:Showed no significant cytotoxicity to 3T3-L1, Caco-2, or HepG2 cells at concentrations up to 40 μM.
Almost completely inhibited lipid droplet formation in differentiated 3T3-L1 adipocytes at 40 μM.
Resulted in lipid droplet formation similar to the uninduced control group in differentiated 3T3-L1 adipocytes at 10 μM.
Increased lipid droplet content as the compound concentration decreased from 40 μM to 10 μM.
