DBO-83 dihydrochloride is a brain-penetrant nicotinic acetylcholine receptor (nAChR) agonist. DBO-83 dihydrochloride activates peripheral nicotinic acetylcholine receptors to evoke non-stimulated ileum contractions. DBO-83 dihydrochloride shows antinociceptive and anti-amnesic activities[1][2].

In Vitro

DBO-83 dihydrochloride (10 µM) shows high selectivity for [3H]-cytisine-labeled nicotinic receptors, inhibiting 85% of binding at 10 µM with no activity against a panel of other receptor types[1].
DBO-83 dihydrochloride constricts guinea pig isolated ileum fragments with an EC50 of 3.05 μM[1].

In Vivo

DBO-83 (1-20 mg/kg; i.p.; 5-7.5 µg per mouse; i.c.v.) dihydrochloride induces dose-dependent, centrally mediated antinociception in the mouse hot-plate tests[1].
DBO-83 (5-20 mg/kg; s.c.) dihydrochloride induces dose-dependent antinociception in the mouse abdominal constriction test[1].
DBO-83 (10-20 mg/kg; i.p.) dihydrochloride induces dose-dependent antinociception in the rat paw-pressure test, with a maximum paw pressure threshold of 104.6 g at 20 mg/kg i.p. measured 15 minutes after administration[1].
DBO-83 (1-10 mg/kg; i.p.) dihydrochloride shows antiamnesic activity in mice[2].

Animal Model: Swiss albino mice in the hot-plate tests (male, 23-30 g)[1]
