GLP-1R prodrug-1 is slowly converted into its active free acid metabolite GLP-1R agonist 45 (HY-187343) in plasma via esterase-mediated hydrolysis. GLP-1R prodrug-1 can be used in the research of type 2 diabetes and obesity.

GLP-1R prodrug-1 (Compound 4) (1 μM; 15 min) shows no activity against GLP-1R in HEK293 cells, while its active metabolite GLP-1R agonist 45 (HY-187343) fully activates GLP-1R with an EC50 of 2.20 nM[1].
GLP-1R prodrug-1 (0.1-1 μM; 1-168 h) undergoes slow, controlled conversion to its active metabolite Compound 4a in pooled plasma[1].
