Heparin disaccharide I-H trisodium (Compound SHS-5) is a heparin-derived disaccharide. Heparin disaccharide I-H trisodium blocks the spontaneous secretion of IL-8 and IL-1β in intestinal epithelial cells at the post-translational stage without reducing mRNA expression levels or intracellular contents. Heparin disaccharide I-H trisodium exhibits dose-dependent anti-inflammatory activity in intestinal epithelial cell models.

In Vitro

Heparin disaccharide I-H (0.01-100 ng/mL; 24 h) dose-dependently inhibits the spontaneous secretion of IL-8 and IL-1β in HT-29 intestinal epithelial cells, with significant inhibitory effects observed starting at the concentration of 0.01 ng/mL[1].
Heparin disaccharide I-H (0.01-100 ng/mL; 24 h) dose-dependently inhibits the spontaneous secretion of IL-8 and IL-1β by Caco-2 intestinal epithelial cells, and exerts a significant inhibitory effect starting at the concentration of 0.1 ng/mL[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

ELISA Assay[1]

Cell Line: HT-29 intestinal epithelial cells
Concentration: 0.01-100 ng/mL
Incubation Time: 24 h
Result: Dose-dependently inhibited spontaneous IL-8 secretion, with significant reduction (P < 0.001) at concentrations ≥0.01 ng/mL.
Dose-dependently inhibited spontaneous IL-1β secretion, with significant reduction (P < 0.001) at concentrations ≥0.01 ng/mL.
Showed varying inhibition intensity relative to other active disaccharides at low concentrations, indicating structure-dependent affinity for cellular targets.

ELISA Assay[1]

Cell Line: Caco-2 intestinal epithelial cells
Concentration: 0.01-100 ng/mL
Incubation Time: 24 h
Result: Dose-dependently inhibited spontaneous IL-8 secretion, with significant reduction (P < 0.001) at concentrations ≥0.1 ng/mL.
Dose-dependently inhibited spontaneous IL-1β secretion, with significant reduction (P < 0.001) at concentrations ≥0.1 ng/mL.
Exhibited a general response pattern matching HT-29 cells, with minor differences in inhibitory kinetics.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
